Next-generation peptides and peptidomimetics

Peptide drug의 단점

1. Poor plasma stability → 오랜 시간동안 남아 target에 binding하여 기능하기 어렵다.
2. Poor membrane permeability → Intracellular target에 binding하기 어렵다.

peptide의 properties를 improve하기 위해서 stabilization하고 non-natural scaffolds로 conversion한 것.**

Peptidomimetics는 보통 4개의 class로 분류 (Class A, B, C, and D)

Class A와 B는 peptide의 성격이 strong함.

Class C와 D는 small molecule을 포함함.

Cyclic peptides, stabilized turns, and loops

• Cyclic peptides
  • cyclic해서 proteases에 덜 노출되어 stability가 높음.
• Stabilized turns and loops
  • Large libraries가 존재해 high-throughout screening으로 다루기 쉬움.
  • 예시) Ensemble Therapeutics (하버드 교수가 2004년에 설립함. Pfizer 등의 기업들도 승인하에 사용 중)

Stabilized $\alpha$-helices and $\beta$-sheets

protein-protein interfaces에서 binding motifs의 $\frac{2}{3}$가 $\alpha$-helices → 따라서 이 구조를 mimic하고 stabilize하는 데에 focus가 되었었음.

• Hydrocarbon-stapled peptides

  • cross-linking and $\alpha$-methylation → helicity를 increase
